Files themselves have to be present in the proper folders at WoW startup, but you can tinker with code while tabbed out, save it, reload and voila! the new code overwrites the old. As long as the file (lua, toc, xml or whatever) is present when you log in, it doesn't matter even if its an empty text file as far as WoW is concerned.
